一个肿瘤细胞是如何建立一个信号通路来便于自身转移的呢?慕尼黑工业大学Klinikum rechts der Isar和 Helmholtz Zentrum München科学家们通过研究结肠癌在这一领域取得了重大发现。他们了解到肿瘤细胞释放趋化因子。在转移性结肠癌细胞中,所释放的趋化因子是CCL2。CCL2趋化因子与血管壁(内皮细胞)相结合,激活相应的受体(CCR2受体)。使得内皮细胞的渗透 性增加,便于肿瘤细胞转移。

Mathias Heikenwälder教授解释说:肿瘤细胞迁移过程中采用一个聪明的伎俩即肿瘤细胞接受健康细胞所发出的信号与内皮细胞相互作用。迄今为止,研究主要集中在肿瘤趋化因子募集巨噬细胞。通过理解趋化因子受体与血管内皮细胞的作用,我们有可能发现一个全新的方法来治疗癌症。

测量趋化因子的数量,可以让我们预测原发肿瘤蔓延到其他器官以及预测患者转移的风险,此外,阻断血管内皮细胞的趋化因子受体CCR2可能为医疗专业人员提供了新的治疗方式。

<br/><strong>Endothelial CCR2 Signaling Induced by Colon Carcinoma Cells Enables Extravasation via the JAK2-Stat5 and p38MAPK Pathway</strong><br/>

Increased expression of the chemokine CCL2 in tumor cells correlates with enhanced metastasis, poor prognosis, and recruitment of CCR2<sup><span style="font-size: x-small;">+</span></sup>Ly6C<sup><span style="font-size: x-small;">hi</span></sup> monocytes. However, the mechanisms driving tumor cell extravasation through the endothelium remain elusive. Here, we describe CCL2 upregulation in metastatic UICC stage IV colon carcinomas and demonstrate that tumor cell-derived CCL2 activates the CCR2<sup><span style="font-size: x-small;">+</span></sup> endothelium to increase vascular permeability in vivo. CCR2 deficiency prevents colon carcinoma extravasation and metastasis. Of note, CCR2 expression on radio-resistant cells or endothelial CCR2 expression restores extravasation and metastasis in <em>Ccr2<sup><img src="http://www.bioon.com/biology/UploadFiles/201207/20120711111925386.gif" alt="" border="0" /><span style="font-size: x-small;">/<img src="http://www.bioon.com/biology/UploadFiles/201207/20120711111925386.gif" alt="" border="0" /></span></sup></em> mice. Reduction of CCR2 expression on myeloid cells decreases but does not prevent metastasis. CCL2-induced vascular permeability and metastasis is dependent on JAK2-Stat5 and p38MAPK signaling. Our study identifies potential targets for treating CCL2-dependent metastasis.
